Dom34 mediates targeting of exogenous RNA in the antiviral OAS/RNase L pathway

In recent years, increasing attention has been focused on the use of synthetic mRNA as a potential new drug for the treatment of infectious diseases, cancer and rare intractable diseases. The mRNA-based therapeutics is expected as a new approach fundamentally different from existing therapy. However, its clinical application is being hindered by the instability of the synthetic mRNA used for the therapeutics. In this study, we have elucidated the molecular mechanism of exogenous mRNA decay and developed a method for stabilizing synthetic mRNA in cells by inhibiting factors involved in the exogenous mRNA decay. Our methodology developed in this study might be the key to opening the way for the development of the mRNA-based therapeutics.
